用传统微技术制作的膜滤光器受到光刻过程的分辨率或曝光光的波长的限制。利用激光的干涉模式可以实现亚微米滤孔。
虽然可以达到分子范围内的孔隙大小,但是产生的孔隙仍在数百纳米量级。聚合物膜过滤器最重要的应用之一是微燃料电池。
间隙过滤器是膜过滤器的替代方案,以实现更小的过滤孔。亚微米的间隙可以通过刻蚀或沉积过程来控制。
虽然微过滤器利用不同大小的物质进行分离,但也可以基于不同特性来实现其他概念,如扩散系数、介电常数、荧光、质量和电荷。
通过介电泳力或光学力,粒子和细胞可以从样品流中进行分类。对要分离的物质进行标记,可以通过外部执行器实现主动分类。闭环控制机制可以基于荧光信号检测物质,并将其切换到收集池。
用于化学分析的微分离器利用了该物质的不同迁移率。不同的迁移率是由不同的质量、电荷或与通道壁上的固定相的相互作用引起的。
这些微量分离器为用于化学和生物分析的便携式和一次性设备提供了巨大的市场潜力。
微过滤器有两个独特的功能:过滤和收集。微过滤器可分离颗粒,用于需要清洁流体的应用,以便进一步加工。微过滤器用于冲洗前处理过程中的废品。
在微反应器中,过滤器用于将微珠保持在反应室中。在其他应用中,过滤后的颗粒也很重要。微过滤器可将颗粒从平均流中分离出来,并将其送至进一步的处理阶段。
根据过滤颗粒的大小范围,过滤可分为传统过滤、超滤和反渗透。传统过滤可过滤10微米以上的颗粒。超滤可处理分子大小在10纳米到10微米的大分子。针对小于10纳米的分子或离子的过滤类型称为反渗透。
根据微过滤器的制造技术和典型的孔径大小,微过滤器可分为膜过滤器和间隙过滤器。
膜过滤器的基本结构是含有过滤孔的薄膜。一般来说,这种膜被称为半透膜,只允许一种成分通过或以较快的速度通过,从而在膜上实现选择性传输.
孔隙可以通过光刻法定义的掩膜蚀刻在膜上。由于光刻工艺的分辨率有限,膜过滤器的孔隙相对较大,约为微米级。
压力、浓度梯度或电势都会影响滤膜上的通量。膜的传输取决于孔的大小。一般来说,传输速率与孔径成正比,而过滤的选择性与孔径成反比。
微机械孔相对较大,大于物质的平均自由部分。通过这种大孔的传输受体积流的支配,分子可以直接通过孔隙。由于孔隙较小,可以假定流动为层流。
如果孔隙小于平均自由通路,但又大到足以进行扩散,那么通过孔隙的传输机制就是扩散。
分子以随机方式通过孔隙。这种随机运动或布朗运动实际上就是一种扩散过程。
如果膜孔足够大,只允许某些物质在膜上扩散,就会发生限制扩散。这种传输机制也称为分子筛选。孔径在0.1至1微米之间时,较大的颗粒无法进入孔内。
这种传输机制在超滤中很常见。如果孔径大小与分子大小相当,例如在大多数聚合物中,物质就会通过体膜扩散。这种传输机制称为溶液扩散或扩散溶解。
间隙式过滤器利用两个结构层之间的间隙作为过滤孔。间隙可以通过表面微加工或体型微加工制造.
例如,在表面微加工中,间隙高度或孔隙大小由作为牺牲层的氧化硅厚度决定,而氧化硅厚度可通过热氧化精确控制。
在体微细加工中,间隙由受控蚀刻工艺决定。这两种微机械加工技术的结合使用可以提高设计的灵活性。
与工业应用相比,许多生物医学应用需要极高的过滤选择性比例。虽然1:102的比例对于行业应用来说就足够了,但生物医学应用可能需要1:104或更高的比例。
这种高选择性只允许孔隙大小的细微变化。通过精确的微加工技术,如光刻和控制牺牲层的生长,微过滤器可以满足这些要求。
由于微过滤器会阻碍水流并造成压力损失,因此要求过滤器上的压降最小。微过滤器最重要的参数是孔隙率,它对压力损失有很大影响。
为了获得最大的孔隙率,膜上的孔应尽可能大、尽可能多。低压损失的另一种解决方案是用横向设计取代膜设计。
横向设计是一种间隙过滤器,它代表了一种流体阻力小的短而宽的通道。
对大孔隙率的要求与所需的坚固性相冲突。较大的孔会降低过滤器的机械强度。因此,应考虑孔隙几何形状对压力损失的影响。
坚固耐用的过滤器是通过间隙设计实现的,间隙由硅块制成,可以承受较高的入口压力。因此,间隙式过滤器可以同时满足低压力损失和高机械强度的要求。
间隙过滤器的优点是孔径小而精确。根据制造方法的不同,间隙过滤器可分为减法间隙过滤器和加法间隙过滤器。
分离器分为主动式和被动式。主动式分离器利用外部能源分离所需细胞和颗粒。所需细胞/颗粒的分离可采用标记法或无标记法。

在基于标记的方法中,首先用荧光团或微珠等标记物对所需分离的细胞进行标记。标记物附着在细胞表面蛋白上。在这种情况下,基于荧光标记的分选和基于微珠的分选方法已经实现。
无标记分离是另一种分离方法,它依赖于细胞物理特性的差异,如大小、弹性、磁感应强度、形状和极化性。
利用荧光探针识别细胞类型是基于荧光标签的分离操作的基础。识别工作由激光检测系统完成。
首先,用荧光标记对细胞进行标记。激光束激发荧光标记,使其发出较长波长的光。显微镜中的外荧光滤光片可检测到该信号,从而识别细胞类型。
然后,通过电泳、介电泳、电渗析、声泳、光学操作、旋转阀和门阀等活性方法,将所需的细胞类型从携带细胞的溶液中分离出来。
在基于微珠的细胞分离中,特定大小、材料和表面结合亲和力的微珠与所需细胞结合。然后借助外场将结合的复合物(细胞珠)分离。
结合细胞受到的力与未结合细胞不同。分离结合细胞可采用磁泳、声泳和电动机制。
下图展示了利用外部磁场从白细胞中分离循环肿瘤细胞(CTC)的过程。CTC最初与磁性微珠结合。在弯曲通道中进行惯性聚焦后,一旦暴露在磁场中,结合的CTC就会偏离主流,并通过磁泳作用被引向收集出口。
无标签分离方法不需要任何标签来识别细胞类型。因此,这种方法需要更简单的样品制备过程,对微流体分离用途非常有吸引力。
这种分离方法依赖于目标细胞固有特性的差异,如暴露于外部能量源(如声学、电场、磁场和光学操作)时的大小或可变形性。重要的是,无需外部能量源的被动方法也能实现无标记分离。
