Fluorine oils, as perfluoroalkane compounds, are widely used as the continuous phase of water-in-oil droplets because of their low viscosity, high boiling point, and good biocompatibility.
What's more, according to the principle of similar solubility, fluorine oil is immiscible with hydrocarbon alkyl compounds, so fluorine oil, as a continuous phase, can not only prepare fluorine oil-in-water droplets, but also fluorine oil-in-acetonitrile, fluorine oil-in-DMF, fluorine oil-in-methanol droplets, and so on, and at the same time, because the contents of the droplets are insoluble in the fluorine oil, so it can realize the encapsulation efficiency of the contents of the droplets of almost 100 percent.

Droplets under fluorescence microscope, it can be seen that only the inside of the droplet glows, and the continuous phase is non-fluorescent, which proves that FC-40 fluorine oil achieves almost 100% encapsulation efficiency for the aqueous solution of the two dyes.

PDMS chips without hydrophobic modification of glass slides, the preparation of water-in-oil droplets will show the phenomenon of water droplets sticking to glass slides, which will affect the generation of droplets.
The PDMS chip successfully prepared uniform and stable oil-in-water droplets after hydrophobic modification of glass slides using hydrophobic modification reagents.
Oil-in-water droplets:
PDMS chips cannot prepare oil-in-water droplets without hydrophilic modification of PDMS.

Droplet breakers:
Extraction of droplet contents is realized after the aqueous phase is separated from the fluorine oil phase by using a droplet breaker to break up the droplets.
